The Waterford community was originally settled by Irish immigrants. About the middle of the 18th century, priests visiting the area were urged to use Shane's Castle for services. Shane's Castle was a rough, cedar log cabin built by three German brothers, Ignatius, Sebastian and Xavier Waas in the mid 1700s. After the demise of the Castle, a wooden church was, built on Church Road, located a short distance to the northeast of the present church building. The present St. Anthony’s Church was built in 1929.

Known as Holy Family Church, the Waterford Catholic community was a mission of St. Joseph Church, Hammonton in the early 1900s. Before 1925 it was transferred to Our Lady of Mt. Carmel, Berlin, then back again to Hammonton in 1940. In the late 40s the Waterford mission, named St. Anthony ‘s in 1929, was attached to the newly-established parish of the Assumption in Atco.

St. Anthony's was named a parish May 23, 1966, with the appointment of Father Pasquale M. DiBuono as pastor. A new rectory was built to house the priests of the parish in 1969.
